Biomembrane Force Probe (BFP) is a highly sensitive single-molecule/cell force spectroscopy technique that enables the quantification of pico-Newton (pN) forces involved in receptor–ligand interactions and cellular mechanotransduction. BFP employs a red blood cell aspirated by a micropipette as an ultrasoft force transducer, with a functionalized microsphere attached to its apex serving as the molecular probe. Upon contact between the probe and a target cell or surface, receptor–ligand binding events induce membrane deformation, which is optically tracked to determine the magnitude, kinetics, and dynamics of the applied force.
